Clinical Significance: The Osmotic Fragility Test measures red blood cell resistance to hemolysis under decreasing saline concentrations, diagnosing hereditary spherocytosis and erythrocyte membrane fragility.
Why It's Done:
Evaluate structural integrity and surface-area-to-volume ratio of red blood cells
Confirm spherocytic RBC hemolysis at higher saline concentrations than normal cells
Differentiate hereditary spherocytosis from autoimmune hemolytic conditions
Guide decisions regarding splenectomy in chronic hemolytic patients
